Effects of Carthamus Semen Extraction on Mecury-Induced Impairment of Organic Cation Transport Function in Renal Tubules




Abstract
This study was undertaken to determine whether Carthamus Semen extraction exerts benefical effect against chemical-induced inhibition of tetraethylammonium (TEA) uptake which is actively secreted by renal pro~mal tubules.
TEA uptake increased as function of incubation time to 120 min. When renal cortical slices were exposed to 0.1 mM HgCl©ü, TEA uptake was significantly inhibited. The inhibition was protected by addition of Carthamus Semen extraction. The benefical effect of Carthamus Semen extraction was dose-dependent over the concentration range of 0.001-0.1%; HgCl©ü (0.1 mM)-induced inhibition of TEA uptake was completely protected by concentrations higher than 0.02% Carthamus Semen extraction. HgCl©ü decreased maximal transport velocity (Vmax) of TEA uptake without a change in Km value, which was completely prevented by 0.05% Carthamus Semen extraction. HgCl©ü and t-butylhydroperoxide increased lipid peroxidation. These changes were prevented by 0.05% Carthamus Semen extraction.
These results suggest that Carthamus Semen extraction exerts protective effect against HgCl©ü-induced impairment of membrane transport function, this effect may be due to by an antioxidant action.
